STRUCTURE ELUCIDATION OF THE METABOLITES OF CH-13584 BY LC/APCI+/MS/MS AND LC/ESI+/MS/MS STUDIES

CH-13584 is a new original antiasthmatic competitor developed in Chinoin.

In vivo studies were carried out in rat and dog to discover the metabolic pathways of the new drug.

The preliminary isolation of the urinary metabolites was accomplished by HPLC resulting in 12 metabolite-fractions. The metabolites less than 1 rel.% were not considered. On line LC-MS/MS methods (daughter-, parent-ion spectra and accurate mass measurements) have been carried out to elucidate the structure of some urinary metabolites of the title compound. Atmospheric Pressure Chemical Ionisation and Electrospray Ionisation were applied as ionisation technics.

Structures were proposed on the basis of the MS data for the main metabolites of several urinary fractions.

Reference compounds with the proposed structures were synthesized and identified by means of the identical MS behaviour and retention time values.

Metabolic pathways were postulated.
